      文章通过GEO数据库获取肝细胞癌样本差异表达基因,利用中药系统药理学数据库及 在线分析平台TCMSP收集双莲方的主要成分及作用靶点,通过Cytoscape构建双莲方中主要成分 的作用靶点与肝细胞癌相关基因的关系网络,预测双莲方治疗肝细胞癌的关键靶点基因,并利用 DAVID进行GO和KEGG富集分析,探讨双莲方治疗肝细胞癌的可能机制。结果表明,从双莲方中 筛选出225种活性成分,与肝细胞癌重合的靶点有28个,主要与肝细胞癌细胞中的激素代谢过程、 蛋白激酶复合物、细胞因子活性等关联,可能通过参与TNF信号通路、P53信号通路、Toll样受体信 号通路等发挥治疗肝细胞癌的潜在作用。研究结果为更深入地研究双莲方对肝细胞癌的作用提 供了理论依据。
英文摘要:
      By obtaining differentially expressed genes in hepatocellular carcinoma (HCC) through GEO database, the main components and targets of Shuanglianfang were collected by TCMSP. The relationship network between the main com⁃ ponents of Shuanglianfang and the related genes of HCC was constructed by Cytoscape. In addition, the key target genes of Shuanglianfang for HCC were obtained. The enrichment of GO and KEGG was analyzed by using DAVID, and the molecu⁃ lar mechanism of Shuanglianfang in the treatment of HCC was explored. A total of 225 active constituents were selected from Shuanglianfang, and 28 targets were coincident with HCC. The targets are mainly related to hormone metabolism, pro⁃ tein kinase complex and cytokine activity in HCC, and involved in TNF signal pathway, P53 signal pathway, and Toll like receptor signal pathway. These may be the potential molecular mechanism of Shuanglianfang in the prevention and treat⁃ ment of HCC. The study provided a theoretical basis for further study on the effect of Shuanglianfang on HCC.
